ssDNA & RAD51 filament

Breast cancer type 2 susceptibility protein

Has eight BRC repeats that bind RAD51. With purified proteins, BRCA2 targets RAD51 to single-stranded DNA, enables it to displace RPA and stabilises the RAD51 filament by blocking ATP hydrolysis. In a BRCA2-mutated ovarian cancer cell line, radiation-induced RAD51 foci were severely impaired, and secondary mutations that restored BRCA2 restored them. [1][2]

Interactions
RAD51, RPA
Clinical relevance
Secondary BRCA2 mutations that restore the protein were found in ovarian cancer cells with acquired cisplatin resistance.
